If I was to put a super on with no Queen excluder and the Queen layed brood in those frames and three weeks before I wanted to take the supers off I put an excluder on ,would they use cells used for brood for honey or is a honey cell a honey cell and a brood cell a brood cell.

You will want to put the excluder on more than 3 weeks prior to extraction. Any drones won't be hatched out at 3 weeks. Workers will hatch at 3 weeks, but you haven't given the bees time to backfill the cells the baby bees just emerged from.

When you put the excluder on, make sure you have the queen below the excluder. Otherwise, she'll keep laying in the honey supers.
