If you are referring to the small plastic "cap" that is showing in your photo, that is pushed into the hole adjacent to it, it holds the cage together. The tube is for release candy, a mini marshmallow should work fine - be sure the queen is inside the cage, first, but don't trap her within the candy.
When I use those for release I put a tooth pick through a pair of windows in one end, then suspend the cage between top bars.
You can also just squeeze them between two frames with the long tube horizontal and on top, that's what they're designed for. Make sure that little attached cap is inserted in the hole tightly (snaps into place), it holds the cage closed. The toothpick trick works good too! Place the cage between two frames of brood, preferably open brood so that numerous nurse bees are available to feed the queen. Mini-marshmallows work great.
I'm going to be installing this queen in a hive that has pretty much NO brood. It's a queenless hive. So, I guess just put it between 2 frames down in the brood nest?
